Ideal Sample Preparation method

Sample preparation for ICP and AA involves converting analyte form so that it is soluble in solvent of interest. This is often achieved by opening up tough chemical bonds in sample by attack of acids and heat. This acid attack results in formation of soluble salts of analyte. Ultimate goal is to convert sample in state presentable to instrument of interest.

Ideal method should be such that,

Brings 100% of sample in solution

Does not allow any analyte to leave the solution

Does not introduces any impurities

Does not brings in any interferences for analysis

Uses acids which are suitable for instrument employed in analysis

Uses acids which are convenient to use

It not very lengthy and can result in high throughput

Fact of life is such method does not exist!

Advantages

l Digestion possible at higher temp than boiling points of acids

l Requires relatively less volumes of Acids

l Use of troublesome acids can be avoided

l Able to trap volatile elements

l Results in very good recoveries

l Environment friendly

Limitations

l Large quantity of sample can not be handled

l Pre-digestion or pre-treatment is often required

l Relatively expensive

l Can not handle large no of samples

l Care is required to avoid cross contamination

l Safety measures are required to deal with large pressures
